1. It will be mounted automatically on boot.
2. It's up to you - if you have small Vita memory card (eg stock 4gb), you can mount SD as ux0 and vita memory card as uma0.

Editing TROPHY.TRP
Does anyone know how properly edit trophies? I've found few apps to extract/repack trophies (TRPViewer, TRPWork) but after translating trophy list and after rebuiding it game always show an error (np-6183-8). Even when i change only one letter in TROP.SFM file. Any ideas how to fix it?

I've never played mission packs but, if they're not counted as mods (so requiring some DLL files), they should be already supported.
